Real radiation yes, but this is supernatural radiation.  I dont think these
mutations come about the same way as real-world mutations.(the DNA is changed, so
over time the body is rebuilt according to a different template)
I see it more as an immediat  physical warping of the body through supernatural
means, with the DNA then changed to make it permanent (and passed on to offspring)

I think they should.  The idea of mutant cyborgs is just too cool to pass up IMO.
And how else do you create a cyborg doomie?  :-)

Would a manitou be affected by Ghost rock radiation?  They are both creations of
teh reckoners, they may be able to ignore it.  As to the AI, that could pose more
of a problem.  I dont know how (real world) computers go with radiation...  I think
they can take quite a bit, especially if built to military specifications.
Certainly they can take more than ordinary flesh.
