The tick would make an "opposed fightin': brawlin'" roll. You would add in the Gunslinger's aptitude in fightin': brawlin' if he is holding a rifle he would gain a defensive bonus of +1 or +2 (I think it says in one of the books if you have them). This would so far make the TN a 9. Since a human is larger than the tick, the tick would get a +6 ( thats what we use. we feel that anything size 4 or higher is about human size so there is no bonus to hit) to hit which would lower the TN to 3. Then since the tick is doing a called shot to the mouth which is similar to a called shot to the eye, the tick would have a -10 penalty which would raise the TN to 13. Since it is a called shot there is no Hit Location to roll. The tick either hits or misses.
